Unveiling another dimension of transcription - G-quadruplex DNA structures are regulatory signposts genome-wide

Transcription is typically studied as transcription factor(s) binding to cognate DNA sequence. We test the intriguing possibility that transcription is regulated by proteins engaging DNA structural motifs instead of sequence. Using the guanine quartet (G-quadruplex) motif we sought to analyse whether DNA structure impacts transcription. This was primarily based on the hypothesis that quadruplex DNA forms must engage trans-factors (proteins) for gene regulatory function. Observations test global DNA-protein interactions along with transcriptional status of human cells to lay a framework for DNA structure-mediated gene regulation.
