The central focus of our chromosome biology lab at IISER Pune is to address the mechanisms of how the nucleus of a cell organizes itself across diverse cell types. The genome has a non-random organization with gene poor chromosomes towards the periphery while gene rich chromosome terrtitories are closer to the nuclear interior. Such a non-random organization also correlates with gene expression, since actively transcribing housekeeping genes predominantly map to the gene rich chromosomes. We have uncovered that lamin proteins are one of the key organizers of the nucleus and the genome, since lamin depletion alters the spatial organization of chromosome territories and gene expression. However, the molecular mechanisms remain unclear.
